Like Gary said – The valves are not spring loaded – by holding the bulb with the arrow pointing up lets gravity aid in the sealing.
Like Frank said – Short story: It is up to you, and your situation.

From what I have read, the fuel filter is near the motor and the squeeze bulb a few inches below that.
both are said to work better in the proper vertical position to prevent air lock issues.
